Jsou zde vysvětleny základní funkce systému a jeho účel a použití v reálném prostředí. Jádrem této bakalářské práce je postup řešení zadaných úkolů v průběhu praxe. Každý úkol obsahuje zadání, postup řešení a závěr. V dalších dvou sekcích jsou popsány využité a chybějicí znalosti. Nakoneci bakalářské práce uvádím závěrečné zhodnocení o probíhané praxiThe content of the thesis is about individual professional practice in company GIRITON Systems s.r.o.. The first part describes the professional focus of the company and the company itself. Next part describes technologies used to accomplish assigned tasks. The third part deals with the information system GIRITON. There are explained basic functions of the system and its purpose and use in the real enviroment. The core of this thesis is about the procedure of solving given tasks during practice. Each task includes a description, process solution and conclusion. In the next two parts I describe used knowledge during the practice, and knowledge gaps. At the end of thesis, I do the final evaluation of the ended practice.460 - Katedra informatikyvýborn

    Inhalation of ZnO nanoparticles: Splice junction expression and alternative splicing in mice

    Despite the wide application of nanomaterials, toxicity studies of nanoparticles (NP) are often limited to in vitro cell models, and the biological impact of NP exposure in mammals has not been thoroughly investigated. Zinc oxide (ZnO) NPs are commonly used in various consumer products. To evaluate the effects of the inhalation of ZnO NP in mice, we studied splice junction expression in the lungs as a proxy to gene expression changes analysis. Female ICR mice were treated with 6.46 x 10(4) and 1.93 x 10(6) NP/cm(3) for 3 days and 3 months, respectively. An analysis of differential expression and alternative splicing events in 298 targets (splice junctions) of 68 genes involved in the processes relevant to the biological effects of ZnO NP was conducted using next-generation sequencing. Three days of exposure resulted in the upregulation of IL-6 and downregulation of BID, GSR, NF-kB2, PTGS2, SLC11A2, and TXNRD1 splice junction expression; 3 months of exposure increased the expression of splice junctions in ALDH3A1, APAF1, BID, CASP3, DHCR7, GCLC, GCLM, GSR, GSS, EHHADH, FAS, HMOX-1, IFN, NF-kB1, NQO-1, PTGS1, PTGS2, RAD51, RIPK2, SRXN1, TRAF6, and TXNRD1. Alternative splicing of TRAF6 and TXNRD1 was induced after 3 days of exposure to 1.93 x 10(6) NP/cm(3). In summary, we observed changes of splice junction expression in genes involved in oxidative stress, apoptosis, immune response, inflammation, and DNA repair, as well as the induction of alternative splicing in genes associated with oxidative stress and inflammation. Our data indicate the potential negative biological effects of ZnO NP inhalation.Web of Science168120019

Tím nemám na mysli popularizaci, zjednodušování, laické převyprávění témat, nýbrž dovednosti kompoziční, schopnosti přehledně látku strukturovat, schopnosti srozumitelně formulovat a správně jazykově vyjádřit myšlenky, postoje, hodnocení atd

In der Form der Schreibmaschinen-Bücher wurden meistens literarische oder wissenschaftliche Werke herausgegeben, die als gedruckte Texte damals in der Tschechoslowakei unzugänglich waren: vergriffene Bücher, Texte der verbotenen oder Exilautoren. Die Exemplare wurden im Freundekreis gratis verschenkt oder geliehen.     Hydrometeor Distribution and Linear Depolarization Ratio in Thunderstorms

    The distribution of hydrometeors in thunderstorms is still under investigation as well as the process of electrification in thunderclouds leading to lightning discharges. One indicator of cloud electrification might be high values of the Linear Depolarization Ratio (LDR) at higher vertical levels. This study focuses on LDR values derived from vertically pointing cloud radars and the distribution of five hydrometeor species during 38 days with thunderstorms which occurred in 2018 and 2019 in Central Europe, close to our radar site. The study shows improved algorithms for de-aliasing, the derivation of vertical air velocity and the classification of hydrometeors in clouds using radar data. The comparison of vertical profiles with observed lightning discharges in the vicinity of the radar site (≤1 km) suggested that cloud radar data can indirectly identify “lightning” areas by high LDR values observed at higher gates due to the alignment of ice crystals, likely because of an intensified electric field in thunderclouds. Simultaneously, the results indicated that at higher gates, there is a mixture of several hydrometeor species, which suggests a well-known electrification process by collisions of hydrometeors

    Optical Absorption Properties in Pentacene/Tetracene Solid Solutions

    Modifying the optical and electronic properties of crystalline organic thin films is of great interest for improving the performance of modern organic semiconductor devices. Therein, the statistical mixing of molecules to form a solid solution provides an opportunity to fine-tune optical and electronic properties. Unfortunately, the diversity of intermolecular interactions renders mixed organic crystals highly complex, and a holistic picture is still lacking. Here, we report a study of the optical absorption properties in solid solutions of pentacene and tetracene, two prototypical organic semiconductors. In the mixtures, the optical properties can be continuously modified by statistical mixing at the molecular level. Comparison with time-dependent density functional theory calculations on occupationally disordered clusters unravels the electronic origin of the low energy optical transitions. The disorder partially relaxes the selection rules, leading to additional optical transitions that manifest as optical broadening. Furthermore, the contribution of diabatic charge-transfer states is modified in the mixtures, reducing the observed splitting in the 0-0 vibronic transition. Additional comparisons with other blended systems generalize our results and indicate that changes in the polarizability of the molecular environment in organic thin-film blends induce shifts in the absorption spectrum.</p
